A preliminary genetic linkage map for the European flat oyster Ostrea eduli ArchiMer

The European flat oyster, Ostrea edulis, occurs naturally from Norway to Morocco in the North-Eastern Atlantic and throughout the Mediterranean. The oyster has been a harvested species for at least 6000 years. However, it successively suffered from over harvesting and then from two protozoan diseases during the 1960s and its production has drastically decreased in Europe from that time. Following two generations of selection for tolerance to one of these diseases (bonamiosis), our aim is to optimize this selection with the assistance of molecular markers, that is to say to implement marker-assisted selection. Oyster productions represent an important part of the french aquaculture. However, diseases are considered as the first limiting factors for the development of this activity around the world. Two exemples point out this problem for the french flat oyster production: Marteiliosis, the disease of the digestive gland, caused by Marteilia refringens, and Bonamiosis, the hemocytic disease, caused by Bonamia ostreae. This report proposes a bibliography analysis of major results obtained for Bonamiosis studies.

The European flat oyster, an endemic species from European coasts, has been classified in the category of "endangered and/or declining species" since 2003. Indeed, the natural beds of this oyster, consumed since ancient times, have gradually been decimated by over-exploitation and by successive emergence of parasitic diseases. Concomitant herpes-like virus infections in hatchery-reared larvae and nursery-cultured spat Crassostrea gigas and Ostrea edulis ArchiMer

Concomitant sporadic high mortalities were reported in France in May 1994 among batches of hatchery-reared larval Pacific oysters Crassostrea gigas and European flat oysters Ostrea edulis in 2 hatcheries, and in June and July 1994 among batches of cultured spat of both species in a shellfish nursery. Histological observation showed the presence of cellular abnormalities in moribund animals. Transmission electron microscopy revealed the presence of herpes-like virus particles in infected larvae and spat of both oyster species. This is the first description of a herpes-like virus infection in larval O.. edulis. Establishment of a genetic map in the European flat oyster Ostrea edulis L. ArchiMer

The flat oyster Ostrea edulis is the species endemic from European coasts, Atlantic and Mediterranean. It has been exploited since Roman times in Europe. But its production decreased from around 20000 tons in the 1950ies to 1500 tons nowadays because of two successive diseases due to the intracellular parasites Marteilia refringens and Bonamia ostreae. Since 1985, Ifremer initiated a program of selection to produce families of oysters tolerant to Bonamia. In this context, a further step is to identify QTLs of resistance to this parasite. Microsatellites development in Ostrea edulis and Mytilus edulis ArchiMer

The European flat oyster Ostrea edulis and the blue mussel Mytilus edulis are both very valuable commercial species across Europe. Despite their economical importance, only a few microsatellite markers have so far been developed in those species. The aim of this study was to develop new microsatellites for O. edulis and M. edulis that could be used for population genetics studies, genetic variability assessment of stocks, parentage analysis or genetic and QTL mapping studies. Population bottleneck and effective size in Bonamia ostreae-resistant populations of Ostrea edulis as inferred by microsatellite markers ArchiMer

Genetic variability at five microsatellite loci was analysed in three hatchery-propagated populations of the flat oyster, Ostrea edulis. These populations were part of a selection programme for resistance to the protozoan parasite Bonamia ostreae and were produced by mass spawns, without control of the genealogy. Evidence for population bottlenecks and inbreeding was sought. A reduction in the number of alleles, mainly due to the loss of rare alleles, was observed in all selected populations, relative to the natural population from which they were derived. Population genetics of the European flat oyster (Ostrea edulis) : from larvae to populations ArchiMer

Nowadays, PCR-based DNA markers allow the study of diversity and differentiation of marine organisms at different stages of their life cycle. Our study of European flat oyster (Ostrea edulis) populations has aimed to describe their spatial and temporal dynamics. First, we analyzed adults sampled over the natural range of the species. An Atlantic/Mediterranean differentiation pattern was observed. Results were congruent between allozymes, microsatellites and mitochondrial DNA and support an isolation by distance model. High variability was observed in the breadth of diversity between populations using a mitochondrial marker, this highlights the small effective population sizes in some locations.
